We are glad to share that this summer, three new papers concerning projects carried at NormandyFlowChem were published:

Process-Intensified Continuous Flow Synthesis of Propofol, OPRD

The first article presents the process intensification of our continuous flow synthesis of Propofol, an anaesthetic used in intensive care units, is now published in Organic Process Research & Development (DOI : https://doi.org/10.1021/acs.oprd.6c00151).

This project was conducted by Antoine Coeuillas, Aurore CEUNINCK, PhD, Laetitia Chausset-Boissarie, Julien Legros, Philippe Jubault and Thomas Poisson within NormandyFlowChem plateforme ofInstitut CARMeNThe purpose was to re-optimize our previous 2021 synthesis, with the aim to scale the process while keeping in mind the various industrial constraints. We could increase significatively both productivity and space-time yield of the synthesis, while discarding non-desirables reagents and solvents for industry, resulting in a more sustainable process.

This work has been made possible thanks to NFC plateforme complete range of flow chemisty equipments: Vapourtec Ltd systems for initial lab development, then Corning Advanced-Flow Reactors (G1) and KHIMOD systems for transposition to higher kilolab/pilot scale.

We also would like to thank Frédéric Pin, Sylvie Dhulut and Thibaud Brégent from ORIL Industrie for GreenScore calculation, and Eric Aubay for the Khimod reactor loan.

Development and Scale-Up of a Regioselective Functionalization of 2-Cyano-3-fluoropyridine in a Flow Reactor, OPRD

The second article presents the development and scale-up of a regioselective functionalization of 2-Cyano-3-fluoropyridine, an interesting intermediate towards APIs, is now published in Organic Process Research & Development (DOI : https://doi.org/10.1021/acs.oprd.6c00156).

This project was conducted by Léna Brassart, Antoine Coeuillas, Thomas Poisson, Julien Legros, Philippe Jubault and Laetitia Chausset-Boissarie within NormandyFlowChem plateforme ofInstitut CARMeNin collaboration with Maxime MANNEVEAU, Philippe Huez and Massimiliano Forcato from the pharmaceutical company Zach – Zambon Chemicals. Le procédé développé consiste en la génération controllée d’un intermédiaire pyridyllithium par déprotonation au LDA en conditions non-cryogéniques puis piégage par l’électrophile désiré.

La chimie en flux a permis de réaliser cette stratégie avec une très bonne sélectivité et une bonne productivité, ce qui a ensuite rendu possible une transposition homothétique de l’échelle du laboratoire (système LowFlow de Corning Advanced-Flow Reactors) à plus grande échelle (système G1). La productivité a ainsi pu être multipliée par 17 sans perte de sélectivité.

Nous sommes également ravis de voir que les deux articles ont été publiés l’un à la suite de l’autre dans le dernier numéro d’OPRD 🙂

Taming the Halogen Dance of Bromo(Thi)azoles through Flow Microreactors, JOC

Le troisième article a pour sujet le contrôle du phénomène de la danse de l’halogène sur des bromo(thi)azoles au moyen de la chimie en flux en continu et son exploitation dans leur fonctionnalisation régiosélective. Il a été publié dans le Journal of Organic Chemistry (DOI : https://doi.org/10.1021/acs.joc.6c00972).

This project was conducted by Baran Ozveren, Antoine Coeuillas, Philippe Jubault, Laetitia Chausset-Boissarie Thomas Poisson and Julien Legros within NormandyFlowChem plateforme ofInstitut CARMeN, en collaboration avec l’entreprise INTEROR.

Les conditions en flux mises au point permettent de favoriser le phénomène de la danse de l’halogène, qui s’appuie sur des échanges brome-lithium entre un intermédiaire déprotonné par du LDA et le substrat de départ, de piéger l’intermédiaire lithié obtenu et d’ainsi arriver plus facilement à divers hétérocycles aromatiques (bi)fonctionnalisés avec une très bonne régioselectivité. Cette stratégie pourrait ainsi servir d’étape-clé dans la synthèses de composés hétéroaromatiques complexes.
